Abstract

Antibody-drug conjugates (ADCs) combine the targeting specificity of a monoclonal antibody with the cytotoxic potency of a small-molecule payload, joined by a chemical linker designed to remain stable in systemic circulation but release the payload once the conjugate is internalized within a target cancer cell. This architecture allows ADCs to deliver drug payloads that would be too toxic for systemic administration on their own, while concentrating their cytotoxic effect on antigen-expressing tumor cells.
